Characteristics of growth, monacolin K and pigment production by Monascus strains on plate culture

|
The production of color pigments and monacolin K by Monascus strains were investigated on agar plates while monitoring cell growth patterns. Thirty five different strains were collected and cultured on potato dextrose yeast extract agar (PDYA), potato dextrose agar (PDA) and malt extract agar medea (MEA) at two different incubation temperature conditions, 25oC and 30oC. The growth rates of most of strains were highest on PDYA medium, and hyphal growth was faster at 30oC than at 25oC. Both red and yellow pigments were highly produced by high pigment-producing strains on agar plate cultures. High pigment-producing strains produced less amount of monacolin K while low pigment-producing strains produced much more monacolin K. Any citrinin was not detected from the monacolin K-producing strains. These results imply that the selection of low pigment-producing strains cultured on agar plates could be applied for primary screening of Monascus strains for preparation of red mold rice.
